The plot revolves around King Leonidas (Gerard Butler), who leads ۳۰۰ Spartans into battle against Persian "god-King" Xerxes (Rodrigo Santoro) and his army of more than ۳۰۰,۰۰۰ soldiers. داستان فیلم تحریفی است از نبرد ترموپیل که در سال ۴۸۰ پیش از میلاد بین سپاه ایران در زمان خشایارشا با سپاه یونان در زمان لئونیداس اتفاق افتاده است. در این داستان، ۳۰۰ نفر از رزم‌آوران اسپارت در برابر سپاه یک میلیونی خشایارشاه مقاومت می‌کنند. این نبرد در تنگهٔ ترموپیل در نزدیکی آتن رخ می‌دهد و حدود سه روز به طول می‌انجامد. ارتش امپراتوری هخامنشی در برابر ۳۰۰ رزم آور اسپارتی در تنگهٔ ترموپیل متوقف می‌شود و تنها زمانی موفق می‌شود آنها را شکست بدهد که در اثر خیانت یک یونانی، راه دیگری برای دور زدن آنها و محاصره اسپارتی‌ها پیدا می‌شود.
